Hi,
I suggest you discuss with your sister inlaw the reason for her opting for separation, is it money or is it freedom or it is housework. Once you know the reason the family can try to change things to make sure the family is together. If not you can plan to have house with 2 floors and can move your brother and his family on different floor this way they will be separate and near also. Win win for everyone.
